Preparing the Stationary Phase

Video. Marking the paper in preparation to spot the sample.

Cut a piece chromatography paper to fit into your development chamber. Mark a point above the solvent level on a piece of scratch paper. Transfer the mark onto the chromatography paper by aligning the bottom of the two pieces of paper and drawing another mark. Do the same thing on the other side of the paper. Then connect the two marks with a pencil line, using a straight edge. It is on this line that all samples will be spotted. Any notes or marks you make on the chromatography paper other than your sample spots must be made in pencil.

For very crude separations, separations can be done using high-grade filter paper as the stationary phase.
